Allosterix LTD is a start up Biotechnology Company founded in 2006. The core competency of Allosterix lies in the design and development novel, allosteric drugs which inhibit enzyme activity.
Enzymes Inhibitors Market
Upwards of 50% of disease pathobiology is attributed to the over-activity or over-expression of an enzyme. For this reason, enzymes inhibitors are key targets for the industry. The main challenge is designing selective and specific inhibitors.
Allosterix technology
Allosterix has developed novel drug design strategy based on multi-disciplinary knowledge and state-of-the-art tools. Quite simply, we design novel inhibitory molecules that are targeted to a located allosteric site. A potential allosteric site is any site on enzyme's surface other than the active site which binding it modulates the enzymes activity. Allosteric sites, unlike active sites, are unique in each enzyme, thus drug selectivity/specificity is inherent in the design of the inhibitor molecule.
Our small molecule inhibitors are made from short peptides, which can be quickly and easily synthesized at low cost, and are ideal candidates for oral delivery.
Allosterix achievements
Target enzyme I – TACE (TNF-ב Converting Enzyme). TACE is a key enzyme involved in various auto immune diseases, including RA (rheumatoid arthritis), IBD (irritable bowel disease) and severe asthma. Due to its billions$/year market potential, it is one of the pharma industry main targets.
Allosterix AL-4-1A1 molecule was the first lead candidate tested in in vivo, mice model. It successfully and significantly (P<0.05) reduced the disease score in the collagen induced arthritis (CIA) mouse model by 30-35%, compared with vehicle control at a dose of only 0.05 mg/kg/day.
Second Target enzyme: BACE-1 (b-Secretase Cleaving Enzyme – 1)
BACE-1 is a key therapeutic target for Alzheimer's disease and the focus of intense research. The company has located an allosteric site on BACE-1 and designed ~400 inhibitors using structure based techniques. Currently, the allosteric inhibitors are being screened for their inhibitor potency in vitro. To date, two lead-compounds have been identified.
Business model
Our core competency lies in locating allosteric sites and designing peptide molecules to bind it. We follow the steps of molecular design and screen and finally in vivo test of the molecule/s in the animal model/s (stage of proof of concept (POC)). After in vivo POC, we are partnering in order to commercialize the lead compound.
